Drebrins (developmentally regulated brain proteins) are cytoplasmic proteins that bind F-actin in the brain and are involved in cell migration, extension of neuronal processes and plasticity of dendrites. There are three isoforms: two embryonic types (E1 and E2); and an adult type (A), generated by alternative RNA splicing from a single Drebrin gene. Drebrins are expressed mainly in brain neurons but are also found in skeletal muscle, heart, placenta, pancreas and kidney. Drebrin has been designated as a marker of the dendritic spine. Decreases in Drebrin levels in the brain have been been associated with Alzheimer's disease and Down syndrome. Recognizes a protein of 130 kDa, identified as Desmoglein-3 (DSG3). This MAb is highly specific to Desmoglein-3 and does not cross-react with other members of the Desmoglein-family. DSG3 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Research has shown that DSG3 has a very high sensitivity (80%) and specificity (100%) in recognizing squamous cell carcinoma (SqCC). Therefore, DSG3 is considered a very important marker for lung SqCC and can be a useful ancillary marker to separate SqCC from other subtypes of lung cancer. Mammaglobin is a 93 amino acid glycoprotein with homology to other secretoglobin-uteroglobin family members. It was originally identified as a breast cancer restricted biomarker by differential screening. Mammaglobin related to secretoglobin family that includes human uteroglobin and lipophilin. Mammaglobin antibody stains cytoplasm of normal breast epithelial cells as well as primary and metastatic breast carcinomas. This antibody recognizes a protein of ~150 kDa, identified as Desmoglein-1 (DSG1). Desmoglein-1 is a member of the desmosomal cadherin family. Desmosomes are intercellular adhering junctions that represent cell surface attachment sites for intermediate filament. Desmocollins and desmogleins are the main desmosomal transmembrane proteins. Desmogleins consist of Dsg1, Dsg2, Dsg3, and Dsg4 isoforms. Within the desmosome, the extracellular domain of desmoglein is essential for calcium dependent heterophilic binding to the desmocollins, whereas the intracellular domain is essential for binding to the desmosomal plaque protein, plakoglobin. Desmoglein 1 is synthesized exclusively in the suprabasal layers. This antibody recognizes a protein of 42 kDa, which is identified as Alpha-methylacyl-CoA Racemase (AMACR), also known as p504S. It is an enzyme that is involved in bile acid biosynthesis and beta-oxidation of branched-chain fatty acids. AMACR is essential in lipid metabolism. It is expressed in cells of premalignant high-grade prostatic intraepithelial neoplasia (HGPIN) and prostate adenocarcinoma. The majority of the carcinoma cells show a distinct granular cytoplasmic staining reaction. AMACR is present at low or undetectable levels in glandular epithelial cells of normal prostate and benign prostatic hyperplasia. A spotty granular cytoplasmic staining is seen in a few cells of the benign glands. AMACR is expressed in normal liver (hepatocytes), kidney (tubular epithelial cells) and gall bladder (epithelial cells). Recognizes a single protein of 33-34 kDa, identified as the prostate specific antigen (PSA). This MAb is highly specific to PSA and stains prostatic secretory and ductal epithelium in both normal and neoplastic tissues. PSA is a chymotrypsin-like serine protease (kallikrein family) exclusively produced by the prostate epithelium, and abundant in seminal fluid. PSA can be detected in the sera of patients with prostatic carcinoma. It is predominantly complexed to a liver-derived serine protease inhibitor, alpha-1-antichymotrypsin (ACT). A higher proportion of serum PSA is complexed to ACT in prostate cancer than in benign prostate hyperplasia. CD45R, also designated CD45 and PTPRC, has been identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. This monoclonal antibody is specific to estrogen receptor alpha (ER alpha) and shows minimal cross-reaction with other members of the family. Expression of NKX2.2 has been found in neuroendocrine tumors of the gut, making it a potential marker for the study of gastrointestinal neuroendocrine tumors. More recently, NKX2.2 protein was identified as a target of EWS-FLI-1, the fusion protein specific to Ewing sarcoma, and was shown to be differentially upregulated in Ewing sarcoma on the basis of array-based gene expression analysis. HHV 8 encodes a latent nuclear antigen (LNA), which is the product of the viral gene orf 73. LNA is capable of forming a complex with retinoblastoma susceptibility gene product, which may be related to its oncogenic activity. HHV8 is associated with three different diseases observed in AIDS patients; kaposi's sarcoma, primary effusion lymphoma (which is a rare type of non-Hodgkin lymphoma affecting the body cavities) and multicentric Castleman's disease.
